A. Physical Examination/Gross Examination 1.Inspection by means of the naked eye or with the use of a hand lens Stain is grayish white to faint yellow in color. In fabrics, the area occupied is depressed. It is usually a map like contour with silvery appearance of the surface. It is hardened with shiny boarder(there are instances that stiffness of the fabric disappear when not properly dried in open air before it is packed.)

The presence of moisture will enhance the growth of bacteria which will destroy the protein constituent of the semen, it will digest the protein and destroy its stiffness. The bacteria will also destroy the albuminous matter and disintegrate the spermatozoa beyond recognition

2. Inspection by means of Ultraviolet light This method is utilized to make small seminal stains or patches visible. Positive = bright bluish fluorescence on clean cloth Cant detect sperm if the clothing is dirty or with blood.

B. Microscopic Examination Identification of the presence of spermatozoa Stain is immersed with water, spermatozoa is transferred in the extracting fluid. A drop of the fluid is dropped and air dried on a slide, stained and examined under microscope

Factors to consider in the microscopic Examination of Sperm cell Chances of locating a spermatozoa is difficult because of the following 1. spermatozoa is bound tightly to the clothing 2. Spermatozoa are brittle when dry thus easily disintegrate 3. when stain is washed and rubbed against other objects 4. when stains are not properly dried, possible of contamination 5. cases of oligospermia, necrospermia and aspermia

1. Florence Test ( presence of Choline Substance) Specimen: Cloth with seminal stains dried thoroughly Procedure: 1. fabric that shows fluorescence is cut from the fabric and is placed in an extracting solution 2. The fabric reagent is withdrawd and the liquid left in the slide depression is treated with a drop of Florence reagent 3. Exam in under LPO and HPO microscope Positive Result= brown Rhombic Crystals

2. Barbiero's test ( test for spermin substance) Specimen: Fresh or dried semen Procedure: 1. Extraction from the stained materials using trichloro acetic acid in a test tube 2. centrifuge, decant the clean liquid and add equal volume of saturated picric acid solution, if precipitate forms heat the preparation until precipitate dissolves 3. remove flame and allow the solution to cool slowly 4. Place the fluid on a slide and treat it with a drop of Barbiero's reagent 5. Exam in under LPO and HPO microscope Positive= definite yellow crystals (leaf shape with irregular boarders) (Spermin picrate)

3. Puramen Test ( Presence of spermin) Procedure: 1. Extraction from stain using saline solution 2. The extract is added with puramen reagent 3. The mixture is refrigerated for few hours Result: Positive Reaction: yellow precipitate 4. If viewed under microscope: precipitate looks to be consisting of a small cross-like crystals of different shape Advantage: most reliable test

4. Acid Phosphate Test (Presence of Acid phosphatase enzyme) The enzyme is secreted by the prostate gland into seminal fluid Reagents used: fast blue dye :Sodium alpha naphthylphosphate Advantage: easy access of enzyme from large fabrics to the collecting material. Moistened filter paper with the reagent then keep it in contact with the fabric suspected to be stained with sperm Positive: purple color
