But, both of these mucous are able to be used as antimicrobial agent. Next stage is bacterial purity test (Gram testing --> Gram negative; Citrate --> negative and Catalase Testing --> positive). Planting E.coli bacteria using the scratch method on EA media. The mucus extraction method used was a steeping method that was modified by soaking the skin and scales and then dried and pounded until smooth and then dissolved with 18% acetone for one day. Testing for antibacterial activity using the diffusion method of paper discs that were modified using filter paper soaked in an antibiotic solution (40% and 60% mucous extract, Cyprofloxacin; NaCl 0.9%) after being dry and the sterile affixed on to EA media. Repeated use of antibiotics in some bacteria causes resistance so that alternative therapies are needed by utilizing natural ingredients, one of which comes from fresh water. Research Goals This study aims to make antimicrobials from Clarias batrachus and Chana striata mucus which have the potential to inhabit the grow of coliform bacteria in Gallus varius L feces, and to determine the minimum inhibitory concentration of Clarias batrachus and Chana striata mucus extracts against Escherichia coli growth. Hypothesis Mucus extract of Clarias batrachus and Chana Striata have antimicrobial activity against the growth of E. coli. The minimum inhibitory concentration of Clarias batrachus and Chana striata mucus extracts on E. Coli growth was 40%. The results of this study indicated that the mucous Clarias batrachus and Chana striata at a concentration of 60% have shown antibacterial activity against the growth of E.coli faecal coliform bacteria originating in the feces of Gallus varius L.
